Lovely map, Mal! Yes, judging by the reference in the top left to General Railway Terminus, it has to date from about 1860. It is irritatingly vague about the link between Swanston St and the road south and, as you say, somewhat conjectural, especially as regards the alignment of the M&HBR. Now, here's another, from Vicnames, which shows Swanston St as extending (now) to approximately the former intersection with Yarra Bank Rd/Batman Avenue and (in deference to what went before?) St K Rd extending over Princes Bridge to that former intersection.

Agreed the attached map from the SLV collection possibly dates from around 1860.

Some of the map is probably conjecture. M&HB railway opened in 1954, Parliament house started construction in 1855 and the government railway terminus at Spencer St was established in 1859.

If I'm correct about some conjecture - then the date may be around 1853.
